Crypt wendtii TC Study

If you have the Crypt wendtii study that was given to us at the AGA in Atlanta, you might notice it leaves a lot out. I got up with Ghazanfar at Outback last night, and we reduced it to simple steps for your (my) ease of use.

MS = Murashige-Skoog

Step 1: Media for initial explant establishment
MS + 1mg/L of BAP + 30g of sugar/L
or
1/2 strength MS + 1mg/L of BAP + 30g of sugar/L

Step 2: Media for multiplication of explant tissue
MS + 4.5mg/L of BAP + 30g of sugar/L

Step 3: Media for growing out plantlets
MS + 0 BAP + 30g of sugar/L

Step 1 can be omitted, but it probably helps to get the explant started on a media with less sugar and BAP.

Step 2 can be repeated as many times as desired before going to step 3 correct?

I believe that is correct. I think doing that is where you get zillions of plants to grow out.

Catch me up on this...Is this what these formulas really mean:

Formula 1: You have one crypt and need to come up with a clean culture (cuz you never know what dirty stuff hangs out in swamps).
Formula 2: You have a clean culture growing--let's subculture it and re-subculture it until you have a zillion plantlets.
Formula 3: Crikey, your basement is filled with test tubes, but nothing growing is bigger than a grain of sand! Subculture a couple of those puppies into new flasks and get them to grow bigger!

Now I have to get more stuff growing immersed!
