I've been getting better about keeping logs of all activity I'm doing with TC. With the amount of jars, media, mixes etc - it gets difficult very quickly to keep everything in memory. Small changes can affect the outcome and its no good to do TC and not be able to replicate/share accomplishments and failures.

They're simple - and designed to be that way otherwise I end up being
lazy about it.

Example Media log:

4/5/2009
Made 1 liter of media - 32 4oz jars ~30ml each
Product used: Caisson Labs MS Product 122 (Instead of logging everything thats in the mix)
1/2 strength MS

Color code: Blue

Added:
1 ml BAP (1 mg/l)
1 ml PPM
30 grams sugar
Adjusted pH to 5.7

Solidified with Agar (Phytotech) 6.5g

Batch code: M001 (You'll see where this comes in handy later)


Example Explant log:

4/6/2009
Initiated VFT (Venus Fly Trap) culture using flower stalks
Dipped in 70% alcohol for 5 seconds
Sterlized in 10% bleach for 20 minutes
Rinsed with sterile water for 10 minutes
Established 5 cultures on M001 media VFT01-05

..and I label each jar with with a code VFT01 - VFT05


Any observations can be logged using the jar code then.
Example:

5/10/2009
VFT01 culture is contaminated with fungus from the explant.
VFT02 culture has 5 shoots


Also, when transferring and explant to different media, you can carry the code though to be able to track something back all the way.

Example:

5/26/2009
VFT01 Transfered to shoot multiplication media - M002


If we all keep half decent logs, we'll have some information to share with others. Obviously, results may vary with each individual, but something is better than nothing.

Which of the agars did you use? The 111 Phytotech agar is suppose to be stronger I have been told. Did you use 6.5g or 7.5?

Nice simple log.

I'm assuming Batch M001 is the 1st batch you made up in March?

The 'logs' above are just examples - not real data, though some of it might be. I'll share some real logs with you guys later tonight if you're interested.

Nah - this is just dummy data. And really, I've been making media for a 3 months now. Just started keeping logs in Feb.

I can post my logs - but use them at your own risk - since I don't know if the results are going to be positive yet.
